Is there a way to make a role assignment temporary? Giving access to a folder on the server that expires, say, at the end of the day.

in my opinion you can schedule the job that can create a user assign and add user to proper role/group and then after processes completed it will delete / remove / disable user.

If the permission needs to be set on operating system folder then you must check under Win2K newsgroups for such snippets, in SQL you can refer to Books online for granting permission on database level.
