    Does the performance tuning wizard really helps to
    improve the access time of database and user experience?
    is there any case study to confirm the above thing.

    Welcome to the forum.
    Yes it help.
    There is no case study (as far I know), but you can find some articles.
    Here is one of them:

    This article was written for sql 2000. The only difference between 2000 and 2005 or 2008 is the name of the tool.
    In 2000 is Index tuning wizard. In 2005 is DTA.
